Evaluating the biological effect of green manure extract in a plant tissue culture system

Abstract

    This study aimed to evaluate the effect of adding green manure and eggshell powder to a tissue culture system used for callus induction and growth in Portulaca oleracea. The experiment was conducted using MS medium prepared with different concentrations of (25, 50, 100) ml/L of green manure extract and (100, 200, 300) g/L of eggshell powder. Callus induction and growth were studied, in addition to measuring fresh and dry weight and protein percentage after (30) and (60) days of inoculation. The results showed that the best response to callus induction was in the treatment to which 100 ml/L of green manure extract and 300 g/L of eggshell powder were added, which led to an increase in the level of other vital indicators. The highest average fresh weight was (3.0, 4.0) at (30, 60) days of age, respectively. As for the protein percentage, its highest value was in the treatment to which 50 ml/L of green manure extract and 200 g/L of eggshell powder were added, reaching (14.08, 17.76) at (30, 60) days of age, respectively.
